Subproject 3: Visualization of interaction between bacteria, tumor and host

The subproject OVGU is primarily focused on the visualization of the cooperation between bacteria, tumor tissue and the host immune system. In order to improve intravital microscopy of bacteria invading tumors in murine model systems, fluorescent labelling of bacteria, tumor and host cells will be optimized.
Thus, it will be possible to analyze the time course of bacterial tumor colonization and oncolysis of a subcutaneously growing solid tumor by intravenously injected E. coli strains developed in the TUD group. In addition to that, it is also important to investigate the impact of the host immune response on this bacterial invasion, for example the recruitment of neutrophil granulocytes. Of special interest will be the analysis of functionally optimized E. coli mutants with oncolytic potential in vivo. Here, time course and effectiveness of tumor destruction by the different newly developed probiotic E. coli mutants will be investigated, as well as the impact of the host immune response in this process.
